Ambrose Island is a fictional island located in the Andaman Sea, Indonesia, and the setting for Shadows in the Water. It is the newest location in HITMAN™ III, but takes place within HITMAN™ 2's timeline. The island consists of rough terrain, being heavily covered with tropical vegetation. Inland, there are high cliffs and deep ravines. Locations include various villages, docks, militia camps, and a cave network. Narrow trails snake through the rocky interior, and several portions of the map are partially underwater.
The island is currently in a lawless state, controlled by various factions of pirates and guerrillas. Despite this, there are civilian villages and even a couple adventurous college students among the inhabitants.

- Before his death, the island was one of Wazir Kale's old ports of call.
- It's one of the three maps not to have Mission Stories, (along with Apex Predator and Untouchable).
- In the water on the upper western beach, there is a barrel partially lying in the ground. Inside that barrel the player can find a pineapple and a sponge. This is a reference to SpongeBob SquarePants.
- The briefing to the mission shows that it's located in the eastern waters of Bengal Bay, and southwest of the coast of Myanmar, which is further corroborated by in-game dialogue.
